Digoxigenin Labeling

Digoxigenin is quite hydrophobic and not water-soluble. Digoxigenin-labeled biopolymers—such as antibodies, proteins, and enzymes—can aggregate and interfere with biopolymer function. AqT® linkers are super-hydrophilic, water-soluble, charge-neutral proprietary materials invented at CellMosaic®. We have modified digoxigenin with an AqT® linker to increase its hydrophilicity and make it readily water-soluble, enabling digoxigenin labeling of biopolymers with minimal or no organic solvent. AqT®-digoxigenin contains multiple hydroxyl (–OH) groups that interact with surrounding water molecules, creating a microenvironment that helps prevent neighboring digoxigenin moieties from stacking or interacting with one another. As a result, biopolymers can be labeled with higher levels of AqT-digoxigenin without altering their properties. This AqT-enhanced H-bond network may also help protect antibodies from enzymatic degradation and support retention of activity.
