Fluorescent Dye Labeling
Commercial fluorescent dyes are often highly hydrophobic. Antibodies labeled with these dyes can aggregate, making it difficult to achieve high dye loading. In addition, even when high loading is achieved, fluorescence quenching can occur due to fluorophore stacking, which reduces the overall fluorescence signal.
AqT® linkers are super-hydrophilic, water-soluble, charge-neutral proprietary biomaterials invented at CellMosaic®. They are designed to label and conjugate biomolecules with fluorescent dyes to improve the performance and sensitivity of dye-labeled bioconjugates. AqT®-dye (a dye modified with AqT® linkers) contains multiple hydroxyl (–OH) groups that interact with surrounding water molecules, creating a microenvironment that helps prevent stacking between neighboring dyes—thereby minimizing fluorescence quenching, even at high dye loading. The AqT®-enhanced hydrogen-bond network also helps protect antibodies from enzymatic degradation and supports retention of biological activity. In addition, it improves water solubility and reduces nonspecific interactions with other proteins and with plastic surfaces. The result is a brighter, more stable fluorescently labeled antibody with improved signal intensity.
AqT®-dye does not introduce ionic groups that can alter antibody properties, as occurs with many commercially modified dyes designed to increase water solubility. The hydrophilicity of the AqT® linker is driven by hydroxyl (–OH) groups and remains charge-neutral.
