AqT® Biotin Site-Specific Antibody Labeling kit is a proprietary biotinylation kit developed at CellMosaic® for obtaining high quality and Fc-region biotinylated antibodies with minimal or no aggregation.
For a detailed description of the AqT® technologies, please see our technology section.
The kit is available in multiple configurations for labeling antibodies (0.1-3 mg) with either one (x1) or three (x3) antibody samples.

Key Features of CM86141 Compared with Typical Commercial Biotin Labeling Kits
AqT® technology enables site-specific biotinylation at reduced Fc-region disulfide bonds, preserving antigen-binding activity while producing more homogeneous, stable, and less aggregation-prone biotinylated antibodies. Compared with conventional antibody biotinylation methods, AqT® biotinylation minimizes changes to antibody properties and provides more consistent labeling results.

|
Labeling Strategy |
Site-specific Fc labeling · Labels reduced Fc-region disulfide bonds (away from the antigen-binding region) · Preserves antibody binding activity and orientation |
Random surface amine labeling · Typically targets lysine residues · Labeling may occur near or within the Fab region · May reduce antibody binding activity |
|
Product Homogeneity |
More homogeneous product with consistent biotin incorporation |
Heterogeneous mixture of differently labeled antibody species |
|
Antibody Properties After Labeling |
Retains native antibody properties · No significant increase in hydrophobicity · No significant increase in apparent MW · Minimal aggregation |
May alter antibody properties · Increased hydrophobicity · Increased apparent MW · Increased risk of aggregation |
|
Linker Technology |
Proprietary AqT® super-hydrophilic, water-soluble, charge-neutral linker |
Zero length biotin or biotin with hydrophobic ethylene- or polyethylene glycol-type linkers |
|
Biotin Loading |
Predictable and consistent · Typical 3–5 biotin molecules per antibody (average » 4) |
Less predictable · Labeling efficiency may vary with reagent quality and storage conditions |
|
Stability of Labeled Antibody |
Excellent Stability · Can be stored frozen or lyophilized without any stabilizers |
High biotin loading may increase aggregation and precipitation |
|
Ease of Use |
Simple Workflow · Complete protocol within ~4 hours · <2 hours hands-on time |
Some rapid-labeling chemistries (e.g., EDC-mediated coupling) may crosslinkantibodies, resulting in large aggregates |

Example Data
The figures below show examples of AqT® biotin site-specific labeled antibodies compared with conventionally biotinylated antibodies. The extent of labeling and the heterogeneity of the labeled antibodies were assessed by hydrophobic interaction chromatography (HIC)-HPLC (Figure 1). Apparent molecular weight and aggregation were evaluated by size-exclusion chromatography (SEC)-HPLC (Figure 2).
Figure 1: Overlay of HIC HPLC profiles of unlabeled antibody (Trastuzumab), Biotin-PEG2 antibody (DOL: 5.2), Biotin-BMCC antibody (DOL: 6.2), AqT® Biotin antibody (DOL: 4.6), and AqT® Biotin antibody (DOL: 6.7).

Figure 2: Overlay of SEC-HPLC chromatograms of the unlabeled antibody (transtuzumab) and AqT® biotin antibody (DOL: 4.6).
AqT® biotinylated trastuzumab show little to no shift in apparent molecular weight and exhibit a hydrodynamic volume similar to that of the native trastuzumab, with no detectable aggregation
