    AqT® Biotin Site-Specific Antibody Labeling kit is a proprietary biotinylation kit developed at CellMosaic® for obtaining high quality and Fc-region biotinylated antibodies with minimal or no aggregation.

    AqueaTether® (AqT®) linkers are proprietary biomaterials developed by CellMosaic for labeling and conjugating biomolecules with highly hydrophobic small molecules. These linkers are designed to improve bioconjugate performance in downstream applications. This kit utilizes AqT® technology to enable site-specific Fc-region labeling, generating high-quality biotinylated antibodies with minimal changes in hydrophobicity and minimal aggregation

    This kit is designed to biotinylate antibodies using AqT® Biotin Maleimide (Scheme 1). The user supplies the antibody, while the kit provides all reagents required for labeling, purification, and biotin quantitation. AqT® Biotin Maleimide is coupled directly to the antibody following reduction of Fc-region heavy-chain disulfide bonds and subsequent alkylation in a single labeling step. The labeled product is then purified to remove any unreacted AqT® Biotin Maleimide. A quick 10-minute biotin assay (reagent and protocol) is also included to determine the biotinylation level immediately after labeling. 

    The kit is available in multiple configurations for labeling antibodies (0.1-3 mg) with either one (x1) or three (x3) antibody samples.

    Applications and Advantageous of Site-Specific AqT® Biotinylated Antibodies

    AqT® biotinylated antibodies can be used in a wide range of biotin/avidin-based applications, including immunoassays such as ELISA, flow cytometry, immunohistochemistry (IHC), immunocytochemistry (ICC), Western blotting, immunoprecipitation (IP)/pull-down assays, biosensor and surface immobilization applications, cell isolation, and magnetic separation. Site-specific labeling at the Fc-region of the antibody helps maintain proper antibody orientation and retain antigen-binding affinity. AqT® biotinylation minimizes nonspecific interactions with proteins and surfaces, improves the stability of the labeled antibody, and provides better sensitivity in streptavidin-based detection systems

    Example Data

    The figures below show examples of AqT® biotin site-specific labeled antibodies compared with conventionally biotinylated antibodies. The extent of labeling and the heterogeneity of the labeled antibodies were assessed by hydrophobic interaction chromatography (HIC)-HPLC (Figure 1). Apparent molecular weight and aggregation were evaluated by size-exclusion chromatography (SEC)-HPLC (Figure 2).

    Conventionally biotinylated antibodies are highly hydrophobic. After labeling trastuzumab with conventional Biotin-PEG2-Maleimide or Biotin-BMCC using the same site-specific reduced thiol labeling chemistry, the peak retention time increased by approximately 0.346 minutes and 0.736 minutes, respectively, compared with the unlabeled antibody (Figure 2). In contrast, AqT® biotinylated trastuzumab with a DOL of 4.6 showed only a slight increase in hydrophobicity, with a retention time shift of just 0.143 minutes at a similar degree of labeling. AqT® biotinylated trastuzumab also exhibited minimal heterogeneity, as indicated by only a slight increase in peak width compared with the unlabeled antibody. Furthermore, increasing the AqT® biotin loading to a DOL of 6.7 did not further increase the hydrophobicity of trastuzumab and resulted in only a slight increase in peak width, indicating only a minimal increase in heterogeneity
