Biotin is highly hydrophobic and only sparingly soluble in water. These properties make it difficult to achieve high biotin loading on a biopolymer. Most conventional biotinylation reagents require significant amounts of organic solvent, which can promote protein aggregation, precipitation, and loss of activity during labeling. CellMosaic’s proprietary AqT® linker converts biotin into a highly water-soluble reagent, enabling high biotin loading with minimal or no organic solvent.
Trastuzumab is a humanized IgG1κ monoclonal antibody that binds to the extracellular domain of human epidermal growth factor receptor 2 (HER2/ErbB2). Trastuzumab is the antibody component of HER2-targeted antibody–drug conjugates such as ado-trastuzumab emtansine and trastuzumab deruxtecan. Biotinylated trastuzumab can be used as a research reagent for HER2 studies and diagnostic assay development. Potential biotin–avidin-based applications include ELISA, flow cytometry, immunohistochemistry (IHC), immunocytochemistry (ICC), immunoprecipitation (IP), pull-down assays, biosensor and surface-immobilization applications, cell isolation, and magnetic separation.

Note: SEC separates the conjugates by apparent MW or size in aqueous solution. In general, higher-MW species elute earlier. SEC is also a useful tool for assessing the level of aggregation in a biopolymer. By comparing the SEC profiles of an unlabeled protein and labeled protein, you can estimate the extent of aggregation in the labeled protein.

Note: HIC separates conjugates based on hydrophobicity. In general, more hydrophilic species elute earlier. HIC is also a useful tool for assessing the degree of heterogeneity in a biopolymer. Comparing the HIC profiles of an unlabeled antibody and a labeled antibody allows changes in the hydrophobicity and heterogeneity of the labeled antibody to be evaluated.
